Arachidonic acid (AA) and other 20 or 22-carbon polyunsaturated fatty acids (PUFAs) are precursors of lipid mediators of inflammation known as eicosanoids. These mediators are critical in disease processes and in regulating normal cell function. Remodeling is important in maintaining homeostasis and in regulating cell function by dictating how PUFAs are converted to lipid mediators of inflammation. Thus, PUFA remodeling is a critical process in the biosynthesis of a multitude of mediators, and understanding this process will unravel better therapeutic targets for controlling inflammatory diseases such as asthma and Alzheimer’s disease. AA metabolism is described in an integrated context linking the remodeling processes with the biosynthesis of mediators and diseases. By following the movement of the substrate (AA), the volume describes how upstream biosynthetic pathways influence the formation of lipid mediators of inflammation, showing the metabolic interrelationship between all AA-derived mediators.

Endothelial dysfunction is broadly defined as a disruption of the balance between vasoactive mediators and a propensity towards an inflammatory state. This volume provides an overview of the fields of endothelial dysfunction and inflammation through the discussion of topics ranging from the molecular biology of activated endothelial cells to the endothelium in inflammatory disease and therapeutic approaches targeting endothelial dysfunction. Topics include: Heterogeneity of the endothelium during inflammation, oxidative stress and endothelial dysfunction, biology and regulation of nitric oxide in inflammatory pathologies, endothelial dysfunction in inflammatory diseases, such as diabetes and atherosclerosis and Clinical methods used to assess endothelial function. This book brings together basic and clinical research to assist the reader in bridging connections from bench-to-bedside. Written by expert researchers in the fields of endothelial biology, inflammation research and clinical science, it serves as a useful reference for academic and industrial researchers, clinicians, and trainees in the medical profession.

Cytokines And Joint Injury

Author : Wim B. van den Berg
ISBN : 9783034878838

The purpose of this volume in the Progress in Inflammation Research series is to provide the biomedical and clinical researcher with a state-of-the-art insight in the role of cytokines in joint inflammation and joint destruction. This is of relevance for better understanding of key processes in diseases such as rheumatoid arthritis (RA) and osteoarthritis (OA). Apart from the impact of old and novel cytokines on joint tissues, the various chapters address the issue of targeted therapy with biological response modifiers and future interventions with carefully designed inhibitors. Spe cial attention is given to elements of synovial cell activation, cell-cell interaction, cytokine interplay as well as mechanisms of cartilage destruction and bone erosion. In addition to an outline of the role of established cytokines, such as TNF, IL-l and IL-6, new information is given on the novel cytokines IL-15, IL-17 and IL-18 and their positioning in the complex cytokine interplay. Cytokine regulation of destructive enzymes, RANKL, the endogenous inhibitor OPG and their crucial roles as central players in joint erosion are highlighted. Together, the chapters provide a complete and balanced view on pivotal cytokines and joint pathology.

This volume contains more than 100 papers presented by leading investigators worldwide at the Ninth International Conference on Prostaglandins and Related Compounds. These presentations cover the entire spectrum of contemporary research on eicosanoids and highlight the applications of this research in the development and therapeutic use of new drugs. Major sections of the book review the most recent advances in the study of phospholipases, cyclooxygenase pathways, lipoxygenase pathways, cytochrome P450 and other oxygenase products, and isoprostanes. Other sections focus on receptors and on the role of eicosanoids in immunology, inflammation, cell differentiation and proliferation, blood cells and the vascular wall, and the respiratory, cardiovascular, and renal systems. Close attention is given to recent research on platelet-activating factor (PAF) and nitric oxide. Also included are extensive updates on ocular prostaglandin research and discussions of the use of prostaglandin E[subscript 1] in the management of erectile dysfunction. Inflammation can be defined as a generalized, nonspecific but beneficial response of tissues to injury. The essential, protective role of inflammation can easily be seen in the life-threatening genetic diseases in which key elements of the inflammatory reaction are missing or defective. The drive to understand the underlying control processes and underlying mechanisms derives, however, more from the pathological perspective than from elucidating defense of the host to injury, and has prompted an enormous effort on the part of the pharmaceutical industry to develop efficient antagonists and inhibitors of the molecules involved in these reactions; the inflammatory mediators. This volume is intended by the authors to provide information on groups of mediators of inflammation in general, and where possible to stimulate interest and comments on the possibilities for participation of these communication molecules, mediators, in the complex but fascinating process of inflammation.
